DNA甲基化(胞嘧啶环第五位碳上共价修饰一个甲基基团)是一种保守、重要的表观遗传修饰,其修饰水平及修饰位点的全基因组特异性分布由DNA甲基化和去甲基化共同决定(Zhu,2009)。拟南芥ROS1(REPRESSOR OF SILENCING 1)是植物中第一个被鉴定的DNA去甲基化酶(Gong等,2002),其通过切除甲基化修饰的胞嘧啶并切开脱嘧啶位点的脱氧核糖核酸链来起始碱基切除修复途径实现DNA去甲基化。近年来,关于ROS1的招募机制及其酶活调控机制逐渐被揭示,但ROS1是否受到翻译后修饰的调控及其机制尚不清楚(Nie 等,2019;Qian 等,2012;Zhang 等,2018)。2020年9月11日,中国科学院分子植物科学卓越创新中心植物逆境生物学研究中心朱健康课题组于Molecular Plant在线发表题为 “SIZ1-mediated SUMOylation of ROS1 Enhances Its Stability and Positively Regulates Active DNA Demethylation in Arabidopsis”的研究论文,首次揭示拟南芥类泛素化E3连接酶SIZ1通过介导ROS1的类泛素化修饰调节ROS1的蛋白稳定性,参与调控DNA主动去甲基化修饰过程。
